Hi, A Open Work Permit is not the same thing as a visa.
The work permit allows one to work for the time stated on the permit.
However the person also needs a valid visa to be in the country, if not from a visa-exempt country.
Therefore the work permit states that this document does not authorize re-entry, because that's what the visa is for. In the end it will depend on the CBSA border agent to let you in again or not, if you decide to leave the country, which is not without risk for inland applicants.
When an inland applicant is denied re-entry the inland application for permanent residence will be considered abandoned and you would need to go through the process all over again from the start. Also at that point since the decision will be made to reject the application from that point on the open work permit will also not be valid anymore.
So if your wife is from a country that needs a visa, I suggest her to apply for a visa as well for the duration of her open work permit just to be safe. That way if she needs to leave the country before the entire process is over, she has the best chances to re-enter Canada at that point.
