Is it okay to pay both the PR processing fee ($475+$75) + ($150) for an initial work permit (change conditions) together
in one Receipt or separately?

Please advice, thank you!

It would probably be okay - but since it looks like you're not going to be paying the $490 Right of Permanent Residence Fee until later in the process, I think (just to be clear) that it's a good idea to pay the $150 for the OWP on a separate receipt. That way there's no way they can confuse things, and you'll have one receipt for $550 in PR fees, one receipt for $150 for the OWP, and a third receipt (later) for final payment on the PR fees of the $490 RPRF.
