My parents were denied usa visa for first time stating, possibility of overstay. Now that their canada visitor visa is going to expire, I'm planning to apply for visitor visa extension, will this usa visa rejection cause problem with canada visitor visa extension.

They have been in canada for 6 months + 2 months extension was approved before
 
You cannot extend a visitor visa. They are applying for a visitor record. Yes a US visa rejection can cause issues when they apply for a new TRV outside of Canada.
 
To improve the chance of renewal of Canada visitor visa, what should I do?
They will need to state the US refusal with reason (IRCC can find it out) and then show absolute ties to return. Seems like they have been in Canada for some time so cannot apply for new TRV until they leave Canada. Then they will show strong ties to return to home country (job, business etc.).
 
They returned to India, they are retired but they have elderly mother to take care of, can I state it in the application?
You can but an elderly mother is not a tie. If they were in Canada for so long then who watched over her? They were out of the country for so long so they cannot be the full time caregiver. Ties are spouse and children under 22. Show that she lives with them, needs 24/7 care and has no other person to care for her. Include medical notes that she needs care too.
