I am the primary applicant for FSW Express Entry and have received the ITA in the 57th draw. I got my MSc degree in Feb 2011 and I started working in Oct 2011. Should I explain the gap between Feb to Sept 2011 in the LOE?

Also, my first job - from Oct 2011 to Dec 2012, does not fit into the NOC code description 2121 in which I have experience from my 2nd and current job. Hence, I had not mentioned about my first job in the work history section in my EE profile. Can I mention it in the LOE that I didn't mention as it was not matching the NOC and hence I didn't claim any points for the first job experience. Or should I just skip it? The time period between my MSc and 2nd job start date is 1.5 years. Will they ignore this gap without any explanation?

You must mention about your first job in personal history section. All gaps must be mentioned - even if it is for even 1 month.
You must explain the gap between graduation and your first job - can mention it as "Unemployed".

Mix and match in NOC codes serves for CRS purposes. It is, thus, not a valid reason for not including it in work history. You can just mention in LoE that you did not include that job in the work history since you did not want to claim any points for it.
