I moved from USA to Canada on Intra Company Transfer Work permit which i believe is LMIA exempt.
Now, in order to claim the 50 points for job offer, is it mandatory to complete 1 year in Canada and be physically present ? If not, what documents are required as proof ? Will the deputation letter be sufficient?

You need one year of experience in Canada on your closed work permit in order to get the points.

No. As you have an open work permit, you need an LMIA to get the points.
