I've been offered a job in Canada, but..

My wife and I currently have our package into the Mississauga office under family sponsorship. However, I've been offered a job with the company I'm currently employed with (Intercompany transfer) at a location on Ontario. If I were to apply for a permit, what effect would there be (if any) on my family visa application?
Could it just process on for my PR status while I work or would I have to start all over as an inland applicant?

No impact to your spousal sponsorship application. It will continue to be processed as is.
