Hello - I recently got approved for PR and have a one time entry stamp to Canada. I currently work in the US on H1B visa. My company is willing to give me a location transfer to Canada, however, my one-time entry expires in couple of months and my relocation process could take over 2 months.
I am thinking of making an entry into Canada and return back to the US. I have relatives who live in Canada and they could probably post me my PR. Is this a safe approach? Would that or has that caused any immigration issues in the past?

For the Canada side of things no issue at all people do soft landings all the time and return to their home country to sort things out. They have their PR cards mailed to a friend or relative who then forward by courier. For sure you need to activate your PR status whilst the COPR is still valid even if you spend just a day in the country.
