First Landing...Does International Transit Count?

Hi, I am about to do my first landing soon from Tokyo and club this with a vacation in US.
I have the following flights with Air Canada
Flight 1. Tokyo to Los Angeles (with transit in Vancouver)
Flight 2. Los Angeles to Vancouver (stay in Vancouver for 1-2 days)
Flight 3. Vancouver to Tokyo

Q1: Now, will this transit in Vancouver (Flight 1) be treated as first landing? If yes, will I be denied entry on Flight 2 as I would have already used my single entry visa and will be allowed entry into Canada only by road after that and not by air?
Q2: I am planning to apply for SIN number (at Service Canada office) after I arrive in Vancouver (Flight 2). I am keeping 1 weekday for this. Is there anything else I need to do during this time? Also, is 1 day enough?
Q3: (Just curious) If transit counts as first landing, can I also pick a flight with a long transit time in Vancouver and complete all formalities along with SIN number. I can thus avoid going back to Vancouver later (Flight 2) and take LA->Tokyo flight only.

Any help will be highly appreciated!

PS: I intend to move to Canada at a later date (hopefully after securing a job).

1. If you have big enough gap between 2 flights. (6 hours at least - why because you never know what would happen there), you can attempt it.
2. The moment your COPR is validated and you will become PR you need your PR card to travel on plane. And it takes some time to get it.
So only if your return flight does not pass through Canada, because to get temporary traveling document can take several weeks.
