I am applying for express entry and I am married and my wife won't be accompanying me to Canada. Is it mandatory for my wife also to give medicals and police verification. If it's not mandatory then what other documents of my wife do I need to submit. As my wife doesn't have a passport yet.

Yes, even if your wife is not accompanying you, her medicals and police verification will also have to be submitted. Moreover, you will have to give clarification to the case officer why your wife is not accompanying you.
