Me and my wife just completed our 2 years in Canada as PR. In the mean time we have been out of the country just for two weeks, and there is little less than 3 years left on our PR cards before they expire. Now since we secured a renewal of the PR, big question is what's next?

Immediately that comes to mind is - two more years of waiting before we can apply for citizenship?

Soooo long!! How soon I should be able to send citizenship application?? Right after I complete 4 years as PR, or may be around 3.5 years just to give a head-start to my citizenship application?

You have to wait until you complete the full four years. If you submit the application a day early, you will be refused. No such thing as a head start.
