I know of at least one case where the applicant used a self declaration and got PPR. His username is
pssid. Having said that this is not a guaranteed approach as it depends on the case officer also.

In your case, I would suggest the following:
1. Get the self declaration with your manager's signature, copy his employee identity card and business card.
2. Also try to see whether your company has advertised any position similar to yours where the duties and responsibilities have been mentioned. It could be either on company website, LinkedIn, any job site etc. Take a printout of the same and attach it with your self declaration.
3. Prepare a Letter of Explanation stating why you cannot get a reference letter from HR/manager and state what proactive action you have taken in this regard, submit the fore mentioned documents along with all employment related documents, offer letter/appointment letter/salary slips/appraisal letter etc etc.
