Please let me know whether we can create express entry for both husband and wife seperately as a family?

Yes you can. If one profile gets invitation , you can either delete other profile or keep it open. There is no harm

This doesn't make sense in my opinion. Why would you want to create 2 profiles? If you have enough points on your profile, you will get the invite - Creating separate profiles simply doesn't make sense. The person who gets the maximum points should be the primary applicant.
 
I would just create 1 profile where I get highest point.

This does make sense for those who do have 400+ points but less than that lucky ITA CRS (450 - 460 :) so as to keep OINP option open.

An applicant should get PT if he has 400. How can 2 profiles help? If an applicant who has 400+ CRS does not get a PT within 3-4 weeks, s/he can consider recreating the profile.
