She cannot sponsor you if the is getting welfare. She does not have to be working but does have to show how, she wouild support you.

It doesn't matter whether you are inside or outside of Canada. Your wife cannot sponsor you if she is on welfare.

Is there is any specific amount that she has to make a month or bi-weekly?
If her family help us? How we suppose to prove that?

If I'm putting my application via Tel-Aviv, it says 40 days stage 1, can I go back to Canada meanwhile?
Can I get get work permit based on stage 1?

There is no minimum amount of income she has to earn.

If her family can help her financially, get a letter from them that states how much they will be contributing. She also needs to stop receiving welfare.

No - you can't get a work permit based on stage 1. This only applies to inland applications. You will be applying outland.
