I am going to send a letter from a colleague who worked with me, as a proof of the duties I have performed, with the appointment letter.

But I do not have the original appointment letter. But I can get a photocopy of the appointment letter from the company I worked and can certify as true copy. Also with the true copy seal.

Will it be enough?? Please help if someone has any idea.....

Yes, it is okay. Reference letter should be original only whereas most of other supporting documents can be sent as certified true copies.

But from one of the places I worked does not issue reference letters with job description. In this case I have the below documents

1) Certified true copy of the appointment letter. Certified with the company true copy seal.
2) Service letter issued by the company with the start and end date of the working period, position held, my name and address.
3) Letter from a colleague with the duties I have performed on a plain latter including the salary, worked hours per week, period of work, work place and location
4) My colleague's official visiting card

What else I need to do?? I can get my colleagues reference letter notarized but is it necessary?

An original letter from your colleague is acceptable if it contains all the required details expected by CIC. No need to notarize anything here, just be sure the original letter is in English or French or an official translation is attached.
