Just recently received PP with VISAS under parental sponsorship and dependents got the visas too from ISB

Well one of them is studying and the other one is not.. they just sent a letter that one of the dependents is not currently studying and is therefore not eligible as a dependent.

All the information was sent. I have a feeling that due to my brothers age they are requesting this as he was only 20 days short of 22 when i applied.

I have just seen an update for status that medicals have been received.

This is very normal if they have any doubts in medicals. The doctor usually asks for 90 days this way if it is viral or something it will go away and they can assess it accordingly. You should not worry about it as i know someone who has been asked to come back in 3 months as well and they...

IF your sibling was a full time student since before 22 years of age until now.. then you have a valid reason to respond to them and advise them of the details.
